CD218b is a single-pass type-1 580 aa glycoprotein. It contains a 19 aa signal peptide, a 342 aa extracellular domain containing 2 Ig-like C2-type segment and 4 potential N-glycosylation, a 21 aa transmembrane domain and a 222 aa cytoplasmic domain which contains a TIR (Toll/IL-1 receptor-like) homologous region. The receptor complex for IL-18 is not yet well characterized. The protein encoded by CD218b is an accessory subunit of the heterodimic receptor for IL-18.

CD218b and CD218a binds togerther as the IL-18 receptor.
|CD218a and CD218b binds IL-18 and mediates the activities of IL-18 through the activation of NF-κB. It is a proinflammatory cytokine that works in concert with IL-12 and promotes Th1 and Th2 immune responses and also induces IFN-γ production and activates NK cells. CD218b is a proinflammatory cytokine that works in concert with IL-12 and promotes both Th1 and Th2 immune responses and induces IFN-γ production and activates NK cells. CD218b enhances the IL-18 binding activity of IL18R1 (IL1RRP), a ligand binding subunit of the IL-18 receptor. Together with IL18R1, it probably belongs to the IL-18 receptor complex but it seems not to interact directly with IL-18. The coexpression of IL18R1 and CD218b is required for the activation of NF-κB and MAPK8 (JNK) in response to IL-18. CD218b is a heterodimer consisting of a ligand-binding α chain, and an associating β chain. Forming a heterodimeric with IL-18Rα enhances IL-18 binding. CD218b is involved in signal transduction.|
