CLEC4A is expressed in dendritic cells, myeloid cells, B cells, HL-60 cells, TNF-α, LPS and down regulated expression at the surface of neutrophils at the protein level. Preferentially, expression is in hematopoietic tissues, peripheral blood leukocytes, moderate levels in spleen, lymph node and bone marrow and low levels in thymus. Expression is also in antigen presenting cells such as monocytes, macrophages and graulocytes. Expression is decreased in dendritic cells by signals inducing its maturation (e.g. CD40 ligand, LPS and TNF-α).

CLEC4A is a single-pass type-2 213 aa glycoprotein. It contains an extracellular domain which contains a stalk domain, 1 C-type carbohydrate recognition domain (CRD), 3 potential N-glycosylation sites and 1 conserved calcium binding site that contains a mannose-binding EPN motif, a transmembrane and a N-terminal cytoplasmic domain which contains a phosphorylated immunoreceptor tyrosine inhibitor motif (ITIM) which is invovled in cellular responses and can bind the SH2 domain of several SH2 containing phosphatases. CLEC4A is closely linked to other C-type lectins on chromosome 12p13 in the natural killer gene complex.
