IL1RL1 is a single-pass type-1 328 aa glycoprotein. It contains an extracellular domain which contains 3 Ig-like C2-type domains and 1 Toll-IL1R (TIR) domain and 9 potential N-linked glycosylation sites, a transmembrane domain and a cytoplasmic domain. IL1R1 type-1 and IL1R2 type-2 forms a cytokine receptor gene cluster in a region mapped to chromosome 2q12.
Alternative splicing yields 3 different isoforms. Isoform B is secreted and isoform C is localized on the plasma membrane.
IL1RL1 has 9 potential N-linked glycosylation sites.

|IL1RL1 is a receptor for IL-33, its stimulation recruits MYD88, IRAK1, IRAK4 and TRAF6, followed by phosphorylation of MAPK3/ERK1 and.or MAPK1/ERK2, MAPK14 and MAPK8. Studies in the mouse suggests that IL1RL1 can be induced by proinflammatory stimuli and may be involved in the function of helper T cells. |

IL1Rl1 does not induce an inflammatory response. IL1RL1 is present on Th2 but not on TH1 T lymphocytes but with neutralization of IL1RL1 enhances TH1 cell responses and inhibits allergic airway inflammation.
