Expression
IL1RL1 is expressed in kidney, lung, placenta, stomach, skeletal muscle, colon and small intestine.  Isoform A is prevalent in the lung, testis, placenta, stomach and colon.  Isoform B is most abundant in the brain, kidney and the liver.  Isoform C is not detected in brain, liver, kidney and skeletal muscle.

Structure
MOLECULAR FAMILY NAME: Belongs to the interleukin 1 receptor family.

IL1RL1 is a single-pass type-1 328 aa glycoprotein.  It contains an extracellular domain which contains 3 Ig-like C2-type domains and 1 Toll-IL1R (TIR) domain and 9 potential N-linked glycosylation sites, a transmembrane domain and a cytoplasmic domain.  IL1R1 type-1 and IL1R2 type-2 forms a cytokine receptor gene cluster in a region mapped to chromosome 2q12.

MOLECULAR MASS

POST-TRANSCRIPTIONAL MODIFICATION

Alternative splicing yields 3 different isoforms.  Isoform B is secreted and isoform C is localized on the plasma membrane.

POST-TRANSLATIONAL MODIFICATION

IL1RL1 has 9 potential N-linked glycosylation sites.

Ligands
LIGANDS AND MOLECULES ASSOCIATED WITH IL1RL1: No information.

Function
IL1RL1 is a receptor for IL-33, its stimulation recruits MYD88, IRAK1, IRAK4 and TRAF6, followed by phosphorylation of MAPK3/ERK1 and.or MAPK1/ERK2, MAPK14 and MAPK8.  Studies in the mouse suggests that IL1RL1 can be induced by proinflammatory stimuli and may be involved in the function of helper T cells.

BIOCHEMICAL ACTIVITY: No information.

DISEASE RELEVANCE AND FUNCTION OF IL1RL1 IN INTACT ANIMAL

IL1Rl1 does not induce an inflammatory response.  IL1RL1  is present on Th2 but not on TH1 T lymphocytes but with neutralization of IL1RL1 enhances TH1 cell responses and inhibits allergic airway inflammation.
