Expression
MPZL1 is expressed with highest levels in heart, placenta, kidney and pancreas.  Isoform 3 is relatively abundant in hemotopoietic tissues and fetal liver.  Isoform 1 and 3 are expressed un CD14-peripheral blood monocytes and pre-B cell progentiors. Isoform appears to be the major isoform in CVD34 promyelocytic and promonocytic cells but during differentiation in monocytic cells, the expression level decreases and that of isoform 1 increases.  Isoform 1 is prominant in stromal cells and to a lesser extent in umbelical vein endothelial cells and erythroid progenitors.  Isoform 2 is expressed in a erythroid progenitor cell line.

Structure

MOLECULAR FAMILY NAME: Belongs to the myelin protein zero family.

MPZL1 is a single-pass type-1 glycoprotein.  It contains an amino-terminal signal sequence, an extracellular domain which contains 1 Ig-like V-type domain and potential N-linked glycosylation sites, a transmembrane domain and a cytoplasmic domain which contains 2 ITIM motifs which is involved in modulation of cellular responses.  The motifs can bind the SH2 domain of several SH2-containing phosphatases.

MPZL1 has potential N-linked glycosylation sites.  Phosphorylated on tyrosine residues occurs upon stimulation with pervanadate and concanavalin (ConA).

Function
MPZL1 is a cell surface receptor which is involved in signal transduction processes.  It recruits SHP-2 to the cell membrane and is a putative substrate.  It is a major receptor to concanavalin (ConA) and is involved in cellular signaling induced by ConA which probably induces Src family tyrosine protein kinases.  Isoform 3 seem to have a dominant negative role as it blocks tyrosine phosphorylation of MPZL1 induced by ConA.  Isoform 1 but not isoform 2 and isoform 3 may be involved in regulation of integrin mediated cell motility.
