MOLECULAR FAMILY NAME: Belong to the dwarfin/SMAD family.

SMAD1 is a 465 aa polypeptide.  It contains 2 conserved functional globular domains, MH1 and MH2, which are connected by a regulatory linker.  SMAD1 belongs to a family of proteins similar to the gene products of the Drosophila gene 'mothers against decapentaplegic' (Mad) and the C. elegans gene Sma.

Function
SMAD proteins are signal transducers and transcriptional modulators that mediate multiple signaling pathways.  It mediates the signals of the bone morphogenetic proteins (BMPs) which are involved in a range of biological activities include cell growth, apoptosis, morphogenesis, development and immune responses.  The phosphorylated form of SMAD1 forms complex with SMAD4 which is important for the transcription regulation.  MH1 binds DNA and MH2 binds membrane receptors for activation, nucleoporins for nuclear translocation and other SMADs and nuclear factors to form transcriptional complexes.  SMAD1 is a target for SMAD-specific E3 ubiquitin ligases, such as SMURF1 and SMURF2, and undergoes ubiquitination and propteasome-mediated degration.
